Diagnosis of Tomato Spotted Wilt Virus (TSWV) in Tomatoes Grown Areas in Isparta and Burdur Provinces, Turkey

dc.descriptionSurveys were carried outin tomato growing areas in Isparta and Burdur provinces in 2014. A total of 78leaf samples were collected from tomato fields in Isparta and Burdur provincesand tested for the presence of Tomato spotted wilt virus by DAS-ELISA using acommercial polyclonal antibody. As a result of DAS-ELISA, all samples werefound to be negative for TSWV. Then these samples were tested for presence TSWVby RT-PCR (Reverse transcription polymerase chain reaction) method amplifyingthe RdRp genes and it was determined that 15 of 78 samples were infected withTSWV. The results that RT-PCR method was found to be more sensitive thanDAS-ELISA for identification of TSWV.
dc.descriptionIsparta ve Burdur illeri domates üretim alanlarında2014 yılında sörvey çalışmaları gerçekleştirilmiştir. Bu sürveyler sırasındadomates üretim alanlarından 78 yaprak örneği toplanmış ve bu örnekler ticaripoliklonal antiserumlar kullanılarak DAS-ELISA yöntemi ile Domates LekeliSolgunluk Virüsü (Tomato Spotted WiltVirus, TSWV) için test edilmiştir. DAS-ELISA testi sonucunda örneklerinhepsi TSWV bakımından negatif bulunmuştur. Daha sonra bu örneklerden RT-PCR(Test Transkripsiyon polimeraz zincir reaksiyonu) yöntemi kullanılarak TSWV’eait RdRp genleri çoğaltılarak 78 örnekten 15 tanesinin enfekteli olduğu tespitedilmiştir. Yapılan bu çalışma TSWV’nün teşhisinde RT-PCR yönteminin DAS-ELISAyönteminden daha hassas olduğunu göstermiştir.
